Abstract: The present study aimed to evaluate antibacterial activity and inhibition mechanism of mixture of betel, gambier and slaked lime in a ratio of 2:1:7 on Staphylococcus epidermidis with measurement on leakage of K+ and Ca2+ using Atomic Absorption Spectrometry technique, leakage of protein and nucleic acid using UV spectrophotometry technique and alteration of bacterial cell wall using scanning electron microscope. The concentrations of mixture used were 0.5, 0.25, 0.125, 0.063, 0.032, 0.016, 0.008 and 0.004%(w/v). The minimal inhibition concentration was about 0.25% (w/v) of mixture. The leakage tests of protein, nucleic acid' K* and ca2* indicated that increasing concentration of mixture was found to enhance its antibacterial activity.
