Abstract: ABSTRACT: The availability of Rhenium-188 as carrier free sodium perrhenate in isotonic saline from a 188W/188Re generator (Knapp et al. 1994; Hashimoto et al. 1996) is popular for a variety of therapeutic applications. Its radiation characteristics (t1/2 = 17 hours, Eβ = 2.12 MeV, Eγ = 155KeV are ideal for theraphy. On the other hand , monoclonal antibodies have shown the potential to serve as selective carriers of radionuclides to specific in-vivo antigens, therefore when we can label antibody monoclonal carcinoembryonic antigen with rhenium-188, this compound could be used for tumor imaging (Radioimmunosintigraphy) and theraphy (Radioimmunotheraphy). In this research radiolabeling carcinoembryonic antigen (CEA) monoclonal antibody was carryout by using directly and indirectly methods. Stannous ion was used to reduce both of rhenium and antibody, before reduce antibody with rhenium was carry out pre reduce antibodies with mercaptoethanol. DTPA was used as bi-functional chelating agent on indirectly methods. Different labelling condition such as pH, incubation time and concentration of reducing agent were all studied for complex formation. The radiochemical purity was determined by using thin layer chromatography and instant thin layer chromatography. The result og yield was found greater than 95% which has fufiled as requirement in radiopharmaceutical kiy for Radioimmunotheraphy.
