Abstract: ABSTRACT INTRODUCTION: Indonesia is the main producer countries of gambier (Uncaria gambir, Roxb) to the world, which is around 80%. The main content of gambier is catechins, but the potential gambier is not widely known, including for drugs, traditionally, Gambier has been used by the people as an anti-microbial, anti-diarrhea, burn treatment and anti-fungal. OBJECTIVES: To know the activity and mechanism of inhibition anti-fungus of catechins on some fungal. MATERIALS AND METHODS: Gambier was obtained from Payakumbuh and partitioned with water and ethyl acetate. Then isolated by column chromatography silica gel using a mixture of chloroform and methanol (4: 1). Antifungal Test of catechins was conducted on C. albicans, S. cerevisiae, F. oxysporum, A. niger and A. flavus. Mechanism of action of catechins on fungi was observed from the leakage of metal ions (Ca2+ and K+) by using AAS. The leakage of proteins and nucleic acids by using UV-VIS spectrophotometry at 260 nm and 280 nm. The changes of fungal cell morphology by Scanning Electron Microscope (SEM). RESULTS AND CONCLUSION: Catechin of Gambier was obtained of around 44, 7%. Anti-fungal activities Test of catechins more potent for S. cerevisiae with MIC values of 12.5 mg / ml. There was an increase the leakage of Ca2+ ion from dose 1 MIC to 2 MIC, namely from 5.96 ppm to 6.36 ppm and an increase in leakage of K+ ion from 88.5 ppm to 89.4 ppm. The increase in absorbance of 1 MIC to 2 MIC at 260 nm from 0.747 to 0.873. Results of the observation of fungal cell morphology by SEM showed the occurrence the plasmolysis and coagulation in cell of fungus. Catechins are not effective against C. albicans, F. oxysporum, A. niger and A. flavus. Keywods: Catechins, Gambier, Antifungal
